The extraction of substantial amounts of high-quality DNA is frequently a challenge in the genetic analysis of plant traits important to agriculture and biotechnology. The extraction of pure, high-quantity DNA from the leaves of 6 Combretum species was investigated using 3 DNA extraction protocols: cetyltrimethylammonium bromide (CTAB), Qiagen and Zymo DNA extraction kits. A modified CTAB method was used for the extraction and purification of sample DNA, while the manufacturers’ instructions were followed for the Qiagen and Zymo kits. The CTAB protocol resulted in DNA with the highest purity reflected by an A260/A280 ratio of 1.9 and a concentration of 900 ng/µL compared to the other methods used. On the other hand, serial dilution of the extraction mixture showed that the more diluted the extraction mixture the lower the concentration of DNA obtained. Among the 6 Combretum species studied, Combretum dolichopetalum Engl. & Diels produced the highest DNA concentration (380 ng/µL), while Combretum bauchiense Hutch. & Dalziel had the highest DNA purity level. The CTAB extraction method produced the best DNA yield and quality for all the Combretum species investigated and can be adopted as a suitable method for Combretum DNA extraction. Similarly, C. dolichopetalum and C. bauchiense can be used for higher DNA yield and quality in further molecular studies.
