Two previously undescribed compounds, xylarinoside (1) and dehydrorhinomilisin G (2), together with five known sesquiterpenes, including hydroheptilidic acid (3), xylaric acid D (4), gliocladic acid (5), 3-acetylgliocladic acid (6), and 14-acetylgliocladic acid (7), were isolated from the wood fungus Xylaria sp. TBRC-BCC29285. Hydrolysis of compound 1 confirmed the absolute configuration of the glucoside unit, while the CD spectrum of compound 2, compared with those of related compounds (rhinomicilin G and pholiotin A), led to an assignment of its absolute configuration. All isolated compounds were evaluated for biological activity, such as antimalarial, anti-TB, antifungal, antibacterial activities, as well as cytotoxicity against cancerous (MCF-7 and NCI-H187) and noncancerous (Vero) cells. Only compound 2 possessed weak anti-TB activity at an MIC value of 50.0 µg/mL.
