Background: Immune Checkpoint Inhibitors have significantly improved outcomes for patients with advanced melanoma. However, 40-60% eventually experience disease progression, highlighting an unmet need for effective treatment options. An anti-CTLA-4/anti-PD-1 combination in PD-1 refractory melanoma achieved an ORR of 28% (90% CI:19%-38%), 6-month PFS of 34% (90% CI: 25%-43%) and a 12-month OS of ~63%1. Evalstotug (BA3071) is a Conditionally Active Biologic (CAB) anti–CTLA-4 monoclonal antibody that minimizes binding at the physiologic pH of normal cells while maximally achieving T-regulatory cell depletion and blocking the interaction of CTLA-4 with its ligands in the low-pH conditions of the tumor microenvironment.
