Fruquintinib or regorafenib? A weighted toxicity score analysis to facilitate better discussions on toxicity and benefits in later-line colorectal cancer treatment

SCSalvatore CoralloFAFrancesco AgustoniAPAnna Pagani

Key Points

  • The aim is to evaluate and compare the toxicity profiles of fruquintinib and regorafenib in later-line treatment for colorectal cancer.
  • Applied a weighted toxicity score to global phase III trials CORRECT and FRESCO-2.
  • Adjusted for toxicity rates in placebo arms to determine net toxicity increase.
  • Fruquintinib increased toxicity by 97.8%, while regorafenib increased it by 85.07%.
  • Regorafenib was linked to a higher risk of dysphonia, oral mucositis, and weight loss.
  • Fruquintinib was associated with a higher risk of hand-foot skin reactions, transaminase elevation, and constipation.

Abstract

In this study, the weighted toxicity score was applied to the global phase III trials CORRECT and FRESCO-2.After adjusting for toxicity rate in the placebo arms, fruquintinib increased toxicity by 97.8% and regorafenib by 85.07%.Regorafenib was associated with a higher risk of dysphonia, oral mucositis, and weight loss, whereas fruquintinib was associated with a higher risk of hand-foot skin reaction, transaminase elevation, and constipation.
