This Viewpoint summarizes the discovery of compound 10d, a natural product-inspired selective ROCK2 inhibitor, whose design was guided by the identification of a novel hydrophobic surface unique to ROCK2. The compound demonstrates compelling efficacy in a mouse model of MC903-induced AD, positioning selective ROCK2 inhibition as a promising strategy for this condition.
