An isolated canine heart preparation was developed which permitted evaluation of the time course of pressure and flow during ventricular diastole. Negative diastolic ventricular pressures were observed after inflow occluded ejections from a wide range of end diastolic volumes. The results are evaluated in the context of recent observations on myocardial ultrastructure and geometry and potential clinical implications are discussed.
