This protocol is for fixation of Aiptasia (Exaiptasia diaphana) larvae and subsequent staining of microtubules, filamentous actin and nuclei. The resulting samples can be imaged under a widefield fluorescence microscope with sufficient image quality to assess basic morphological features such as size, gastric cavity and pharynx, cilia and apical tuft. For highest quality images, a confocal laser-scanning microscope can be used.
