5-Fluorouracil is a uracil analog that has been used for over 65 years to treat a wide variety of neoplasms and diseases. It does so by disrupting normal nucleotide synthesis of both DNA and RNA, leading to tumor cell death 1. This review aims to discuss the history of this medication, its mechanisms of action, and address the need for further research into increasing efficacy and preventing tumor resistance.
