Evaluation shows bismuth acetate paper effectively detects hydrogen sulfide, implying a safer alternative to lead acetate methods.
Hydrogen sulfide (H₂S) detection is crucial in environmental monitoring, clinical diagnostics, and industrial safety. Traditional colorimetric detection using lead acetate-based paper chromatography assay provides rapid, inexpensive, and high throughput assay to detect and measure hydrogen sulfide levels. However, due to toxicity of lead compounds additional safe handling and disposal requirements make it less attractive for common use. Herein, we evaluate bismuth acetate and copper acetate paper as safer alternatives. Paper preparation parameters, including drying methods and humidity, were optimized. Bismuth acetate papers demonstrated consistent homogeneity and reproducibility, comparable to lead acetate papers and superior to copper acetate papers. The detection range for bismuth acetate paper was comparable to lead acetate, outperforming copper acetate paper. Practical applicability was validated through detecting biologically generated H₂S from HepG2 cells, confirming the paper's suitability for biological assays. These findings support bismuth acetate paper as a safe, effective alternative for high-throughput, colorimetric detection of hydrogen sulfide.
