Observational analysis reveals changes in glioma perfusion metrics in mice, indicating vasculature development.
Motivation: Characterize glioma tumor mice perfusion during progression using BOLD-DSC in 9.4T. Goal(s): Investigate perfusion parameters including AUC, peak and FWHM in GL261 glioma mice. Approach: Using short 5s gas challenge (100% N2 / 0% O2) to manipulate the change in deoxyhemoglobin concentration as an endogenous contrast agent to assess perfusion metrics. Results: BOLD-DSC is a sensitive and repeatable method to evaluate tumor progression. Changes of perfusion, specifically contributed by large vessels, were observed during tumor progression. Excessively prolonged transit time is the consequence of inefficient tumor vasculature and provide an excellent contrast for tumor localization as well as sensitive biomarker of severe progression. Impact: BOLD-DSC is used as a sensitive and repeatable tool to longitudinal investigate glioma tumor perfusion during development, giving an insight into the vasculature change by directly observing AUC, peak and FWHM.
