Genetic examination identifies species substitution in canned salmon products, highlighting DNA extraction effectiveness.
A genetic examination of canned pink (Oncorhynchus gorbuscha), chum (Oncorhynchus keta), and sockeye (Oncorhynchus nerka) salmon was conducted using real-time PCR. Two DNA extraction methods were compared: proteinase K digestion and alkaline lysis. The extracted mitochondrial DNA was analyzed using real-time PCR. The effectiveness of DNA analysis de-pends on the sample size and its representativeness. DNA of chum salmon was detected in a sample labeled as sockeye salmon, indicating a possible substitution.
