Observational analysis shows polyamine depletion does not prevent tumor growth in Ewing sarcoma, indicating ferroptosis is involved.
Supplementary Figure S5: 1% DFMO does not prevent tumor growth. Fragments of the JHHESX3 PDX were implanted either in the pretibial space (orthotopic; panel A) or subcutaneously (B) in 10 NSG mice, and mice were randomly assigned to regular drinking water or water supplemented with 1% DFMO (w/v). Tumor size was assessed twice weekly. (C) Fragments of the JHHESX3 PDX were implanted orthotopically in 15 NSG mice. When tumor/leg circumference reached 30 mm, the tumor-bearing leg was amputated. Five mice were given regular drinking water throughout the experiment, 5 were given water supplemented with 1% DFMO (w/v) beginning at the time of tumor engraftment, and 5 were given water supplemented with 1% DFMO (w/v) upon recovery from amputation. Mice were followed until body condition score dropped to 2 or lower and then were euthanized. Neither treatment prolonged post-amputation survival.
