Reactive oxygen species formed in living organisms from various factors, cause to DNA damage, cytotoxicity and cell death. This study aims to investigate protective effect of two varieties of olive leaves extract against oxidative DNA damage induced by Fenton reaction. Gemlik variety (black olive) extract was found to have a slightly stronger protective effect than Domat (green olive) variety extract against oxidative DNA damage. Gemlik variety extract until did not show toxic effect on healthy liver cell line (AML12), it had shown cytotoxic effect on liver cancer cell line (Hep3B). However, Domat variety extract showed cytotoxic effect on Hep3B only at 24-hour incubation time. While both extracts contained the most oleuropein, chlorogenic acid and protocatechuic acid, the Gemlik variety extract contained two times more rutin and luteolin than the Domat variety extract. These results suggest that the higher concentrations of these compounds in the Gemlik variety may be attributed to these findings.
