aims: To study the effect of early intervention with puerarin on High-Speed T2-Corrected Multiecho Acquisition at 1H MR Spectroscopy(HISTO) quantification of hepatic hepatic fat signal fraction in intrauterine growth restricted(IUGR)rats. background: Proton Magnetic Resonance Spectroscopy (1H-MRS) measurement of hepatic fat content in intrauterine growth restriction rats has rarely been reported objective: To study the effect of early intervention with puerarin on High-Speed T2-Corrected Multiecho Acquisition at 1H MR Spectroscopy(HISTO) quantification of hepatic hepatic fat signal fraction in intrauterine growth restricted(IUGR)rats. method: Pregnant mice were divided into the control, IUGR, intervention groups. Intervention and IUGR dams were fed a low-protein diet during pregnancy, whereas dams in the control group were fed a normal diet. The pups in the intervention group were given a unilateral intraperitoneal injection of 50 mg/kg/d puerarin 1 week post birth. Male rats were selected and weighed, the body length and waist circumference were measured at 3, 8 and 12 weeks post birth respectively. These male pups also received conventional magnetic resonance imaging and HISTO using a 3.0 T whole-body MR scanner at 3 ,8 and 12 weeks post birth, hepatic fat signal fraction(HFF) was calculated afterward. result: Significantly lower body weight,BMI, and body length at 3 weeks were observed in newborn pups of IUGR and Intervention Rats compared to pups of control rats.Significant differences in the body weight, body mass index (BMI), waist circumference and HFC were observed between IUGR and control rats at 8 and 12 weeks post birth.Meanwhile, significantly higher HFC was observed between the treatment and control rats at 8 and 12 weeks post birth. Significantly difference in HFF was also observed between the IUGR and treatment rats at 12 weeks post birth, nevertheless there was no significant difference at 8 weeks post birth. conclusion: Early postnatal puerarin intervention has an important effect on liver fat signal fraction and may help reverse adult obesity in IUGR rats. other: HISTO allows the assessment of hepatic lipid
