In the recent five years, the intravesical instillation of EGF in interstitial cystitis (IC) was barely in progress. This study was the first one discussing the efficacy of intravesical instillation of EGF in rats with LPS-induced IC. METHODS: There were 15 female Sprague-Dawley rats used in this experiment, and they were divided into sham control, IC, and treatment group. The commercialized EGF product NewEpi (GoodCare Biotech, Taipei, Taiwan) was utilized. We employed 150 μl LPS to induce IC in rats, and then each rat in treatment group was infused with 400 սl NewEpi solution on day 0, day 7, and day 14 after IC induction. On day 21, the urinary bladder (UB) tissues were retrieved to analyze the inflammatory cytokines and oxidative markers. Tukey's HSD test was adopted to determine the differences among multiple treatments in one-way ANOVA. RESULTS: After instilling EGF, the urothelial inflammation was lessened, the fibrosis was ameliorated, the mast cells' infiltration was improved, and the urothelial denudation was recovered (Figure 1). In our results, EGF could remarkably down-regulate the pro-inflammatory and pro-fibrotic cytokines such as TNF-α, IL-6, and IL-1β (Figure 2A). In oxidative stress assessment, after instilling EGF, the free radicals' scavengers, GSH and SOD, were all significantly up-regulated, and the lipid peroxidation product MDA was lowered in UB (Figure 2B). CONCLUSIONS: By instilling EGF in rats with LPS-induced IC, urothelial inflammation and fibrosis were lessened. The mechanisms were postulated that it could down-regulate inflammation-related cytokines and up-regulate free radicals' scavengers. EGF could effectively lower the peroxidation of UB, and it is worth being studied in human clinical trial.
