Noncoding RNAs are emerging as crucial players in regulation of gene expression, cell signaling, and metabolism. Studies from our laboratory demonstrated that lncRNA HOTAIR regulates inflammatory response in macrophages and modulates cytokine expression via regulation of NF-kB activation. Additionally, we also discovered a series of novel lncRNA modulators of inflammation in mice and human, termed as Long-noncoding inflammation associated RNA, LinfRNA). Some of these hLinfRNAs (such as hLinfRNA1) control NF-kB signaling and some others regulate JAK/STAT signaling. Additionally, hLinfRNAs also regulate glucose metabolism and lipid homeostasis in macrophages during inflammatory response. Here, we will present our recent studies in functional characterization of selected inflammatory lncRNAs in regulation of cytokine expression, NF-kB signaling, and their roles in glucose/lipid metabolism during inflammation and macrophage activation. Overall, our studies demonstrated that lncRNAs are novel regulator of inflammation and metabolism and hence have potential towards discovery of novel biomarkers and therapeutic targets for inflammatory human disease.
