Research shows antigen presentation enhances antibody secretion in B cells, suggesting new therapeutic pathways for autoimmune diseases.
Description Although B cells are essential for defense and long-term immunity against invading pathogens, their hyperactivity can contribute to the development and maintenance of autoimmune diseases, such as Systemic Lupus Erythematosus (SLE). There is limited data linking antibody secretion to specific phenotype, genetics, and antigen presentation. We are assessing whether nanovials, modified hydrogel particles, can enable singe-cell analysis by capturing cells and specific secreted products for further downstream analysis. Our research determined the media that maximizes IgG secretion, successfully loaded cells into nanovials, and detected IgG secretion. This workflow will allow us to isolate antigen-specific antibody secreting B cells to identify surface markers and phenotype contributing to therapeutic development to target specific pathogenic B cells.
