This work demonstrates enhanced immune monitoring in cancer and autoimmune diseases using a 30-color spectral flow cytometry panel.
Description OMIP Panels (Optimized Multiparameter Immunofluorescence Panels) are pre-designed, highly optimized flow cytometry antibody panels that enable the simultaneous detection of multiple immune cell markers in a single sample. OMIP panels are designed to optimize fluorochrome choices, antibody combinations, and compensation strategies, ensuring that they can work effectively with various flow cytometry instruments. This study focuses on the re-optimization and validation of the established OMIP-104 spectral panel on the new CytoFLEX mosaic 88 Spectral Detection Module, which provides spectral capabilities for our CytoFLEX LX instrument. The primary focus is on its ability to analyze complex cell populations with enhanced precision and sensitivity. The optimization process involved adjusting antibody titrations, spectral settings, and unmixing protocols to minimize spectral overlap and maximize signal clarity. The use of the new CytoFLEX mosaic Spectral Detection Module allowed for the simultaneous detection of multiple markers, greater resolution and deeper insights into the immune system. By leveraging the capabilities of the new spectral module, this OMIP panel underscores the importance of OMIPs for reproducibility across different analyzer platforms with minimal effort. This work confirms the potential of spectral flow cytometry to transform immune monitoring, providing a powerful tool for researchers investigating cancer, autoimmune diseases, infections, and immunotherapies.
