Synthesis and structural characterization of a benzimidazolone compound shows promising antidiabetic activity and good toxicity properties.
A new compound was synthesized from benzimidazolone and 2‐chloro‐ N ‐phenylacetamide in excellent yield. This compound was identified based on their spectral data analysis ( 1 H NMR, 13 C NMR, and FTIR). The structure of this benzimidazolone was confirmed by X‐ray diffraction analysis. The density functional theory (DFT) method with the B3LYP functional and the 6–31++G(3d,2p) basis set was used to optimize the studied molecule. The frontier molecular orbitals, the highest occupied molecular orbital and the lowest unoccupied molecular orbital, have been determined, and the energy gap between these two orbitals has been calculated. To analyze the chemical reactivity of the molecule, the main reactivity indices, the condensed Fukui functions as well as the molecular electrostatic potential were calculated and analyzed. Fingerprint plots were used to explore intermolecular interactions, we also used the reduced density gradient technique to study a variety of noncovalent interactions inside molecular structure. In addition, docking investigations of this product revealed important interactions with the active sites of both N‐terminal human maltase‐ glucoamylase, α‐glucosidase, and α‐amylase. The analysis revealed that the studied compound had lower binding energies ranging between −8.4 and −8.8 kcal/mol. and formed stable complexes at the protein's substrate‐binding site at 100 ns molecular dynamics simulation. The pharmacokinetic and toxicity properties of synthesized benzimidazole were also examined and revealed good drug‐like qualities.
