This technical brief dissects US Patent 11,219,685, a CD40-targeted intranasal MERS-CoV vaccine and identifies critical translational vulnerabilities, including reliance on murine CD40L, narrow intranasal formulation constraints, consensus S1 antigen limitations, and Ad5 vector liabilities. Drawing on large-scale genomic surveillance and structural virology, our analysis reframes these gaps not as dead ends, but as actionable opportunities for de-risked, human-compatible redesigns with clear pathways to regulatory and commercial viability in the MENA region and beyond.
