In a synthesis approach, azoidine intermediates enable production of triamines in a drug development context, highlighting potential for diverse applications.
The synthesis of pactamycin natural products and κ-opioid receptors, along with their analogues, which feature three contiguous cis-trans chiral carbon-nitrogen centers, holds substantial research value in drug development. Notably, we have successfully developed a concise and efficient methodology for constructing a molecular framework with three contiguous carbon-nitrogen stereocenters utilizing 1,2-nitrogen migration and the stereoselective addition of nitrogen nucleophiles from aziridine intermediate. This transformation demonstrates excellent substrate generality and broad applicability, providing a solid foundational basis for the synthesis of pactamycin natural products, κ-opioid receptors and other aminocyclitol-containing antibiotics.
