Radical enzymatic peptide cyclization in natural product biosynthesis

ZYZiwei YaoBMBrandon I. Morinaka

Key Points

  • The paper aims to discuss the role of radical enzymes in the cyclization of cyclic peptide natural products.
  • Review of existing literature on radical enzymes and cyclic peptides.
  • Analysis of different enzyme families involved in cyclization.
  • Discussion of their significance in drug discovery.
  • Radical enzymes produce a variety of unique cyclic peptides.
  • Different enzyme families contribute to diverse macrocycle formations.
  • Cyclic peptides are emphasized as important scaffolds for future drug development.

Abstract

Cyclic peptide natural products are privileged scaffolds in drug discovery. Here we review cyclization by radical enzymes which create diverse and unique macrocycles and comprise several distinct enzyme families.
