Activity-based sensing and labeling (ABSL) has emerged as a powerful platform for the selective detection of transient biological analytes with high sensitivity and spatial fidelity. Here, we highlight emerging molecular design strategies that leverage ABSL for advanced investigations of critical cellular signaling processes. We summarize key biological insights resulting from ABSL of enzyme activity, reactive oxygen and nitrogen species (ROS/RNS), and labile transition-metal pools.
