Key Points

  • The aim is to evaluate the role of microRNA-1 in regulating metabolism during skeletal muscle hypertrophy.
  • Review of current literature on microRNA-1 and its role in muscle physiology
  • Analysis of how mechanical overload affects miR-1 expression
  • Investigation of metabolic pathways linked to hypertrophic adaptation
  • Downregulation of miR-1 is a consistent feature across various species during muscle hypertrophy.
  • Reduced miR-1 levels contribute to metabolic changes that support muscle growth.
  • Evidence suggests miR-1 serves as a master regulator coordinating these processes.

Abstract

Downregulation of microRNA-1 (miR-1), the most abundant muscle-enriched microRNA, represents a conserved hallmark of skeletal muscle hypertrophy across species. We propose that mechanical overload-induced reduction in miR-1 expression drives metabolic reprogramming critical for hypertrophic adaptation. This review explores emerging evidence establishing miR-1 as a master regulator of metabolism that governs skeletal muscle growth. Summary: microRNA-1 downregulation during mechanical overload enables skeletal muscle hypertrophy through metabolic reprogramming.
