Computational analysis identifies promising 1H-3-indazole derivatives for cancer treatment, suggesting further investigation is needed.
Cancer remains a major global health burden requiring the development of novel therapeutic scaffolds with improved efficacy and reduced toxicity. Indazole-based heterocycles have emerged as attractive pharmacophores due to their kinase inhibitory and multi-target anticancer properties. The current work uses structure-based computational techniques to analyse four 1H-3-indazole derivatives. Molecular docking was carried out against twelve cancer-related protein targets. Drug-likeness and ADME properties were predicted using Swiss ADME, and toxicity was assessed with Pro tox-III. All compounds met Lipinski's Rule of Five and Veber's Criteria. Ligand-3 consistently demonstrated strong binding to numerous targets, with docking scores ranging from -7.4 to -13.0 kcal/mol. Toxicity prediction classed the compounds as Clas-IV with adequate safety margins. These data indicate that 1H-3- indazolederivatives, particularly Ligand-3, could be intriguing multi-target anticancer lead candidates that require further experimental validation.
