This working paper demonstrates eleven mechanisms of addiction in relation to oxytocin in a neurobiological framework, suggesting new prevention strategies.
What if the most effective approach to preventing and treating addiction lies not in pharmacological intervention — but in strengthening the neurobiological system that underlies human social bonding? This working paper proposes a unifying answer: the distinction between exogenous oxytocin (pharmacologically administered) and endogenous oxytocin (produced through genuine social bonding) may be the key to resolving this paradox. The Oxytocin Multi-Target Model (OXT-MTM) organises eleven documented mechanisms through which oxytocin modulates addiction-related processes, systematically mapped onto the three stages of the Koob and Volkow (2016) addiction cycle — binge/intoxication, withdrawal/negative affect, and preoccupation/anticipation. Unlike previous reviews that focus on individual mechanisms or specific substances, the OXT-MTM provides the first numbered, stage-mapped integration of the full mechanistic portfolio, building on the foundational work of Bowen and Neumann (2017). Key contributions: Eleven mechanisms across three stages: From dopamine blockade in the nucleus accumbens (Peters et al., 2017) through HPA axis modulation, BNST rebalancing, and craving reduction to epigenetic regulation of OXTR expression (Proskynitopoulos et al., 2025) — organised within the established Koob and Volkow (2016) framework. The endogenous/exogenous distinction as explanatory principle: Intranasal OXT shows inconsistent clinical results (16/17 trials with high bias risk; Mellentin et al., 2023). The model argues that oxytocin is an inherently social neuropeptide whose effects evolved within the context of social interaction — pharmacological administration without an accompanying social bonding context contradicts the neurobiological operating logic of the system. Faith-based bonding as an OXT activation pathway: Attachment research has established that God can function as a noncorporeal attachment figure (Granqvist, 2020). Endogenous oxytocin levels were approximately twice as high in individuals reporting spiritually transformative experiences (Kelsch et al., 2013). Lambert et al. (2010) demonstrated that personal prayer alone reduced alcohol consumption by ~50% in a causal design (N = 1,758). Prevention from birth, not just in adolescence: A four-stage developmental safety net model proposes interventions from birth (midwifery, secure bonding) through preschool (parental training), primary school (family resilience), to pre-adolescence (ages 8–10, booster before typical substance initiation at 12–14). Sex-specific dual-mechanism architecture: The dopaminergic pathway operates primarily in males (Peters et al., 2017; Logan et al., 2022), while the glutamatergic pathway functions in both sexes — specifying rather than undermining the model. Contribution to Sustainable Development: This research directly addresses UN SDG 3.5 (substance abuse prevention) and SDG 3.a (tobacco control) by translating convergent neurobiology into a theoretically grounded framework for prevention programme design. The model generates testable predictions for human research and offers a structured roadmap for piloting bonding-based interventions with accompanying scientific evaluation.
