Research of EA on mitophagy in the midbrain substantia nigra of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine mice

Abstract
目的 观察电针对帕金森病(PD)小鼠中脑黑质线粒体自噬的影响,探讨电针治疗PD机制。 方法 小鼠随机分为正常组、模型组、电针组、模型+3-MA组、电针+3-MA组,12只/组。模型组小鼠腹腔注射1-甲基-4-苯基-1,2,3,6-四氢吡啶(MPTP)(30 mg/kg)7天。造模成功后,电针组小鼠每日电针20 min,连续5天;模型+3-MA组小鼠腹腔注射3-MA(25 mg/kg)5天;电针+3-MA组小鼠腹腔注射3-甲基腺嘌呤(3-MA),2h后电针20 min,连续5天。旷场观察行为变化;组化及荧光测TH、α-syn、LC3;电镜观察黑质超微结构;Western Blot和RT-PCR检测TH、α-syn、PINK1、Parkin、P62、Beclin-1、LC3蛋白和基因表达。 结果 与正常组比较,模型组运动功能障碍(PPPPPPPP 结论 电针通过增强线粒体自噬改善PD小鼠运动障碍,减少α-syn聚集,增加TH,恢复线粒体形态,保护DA。
