Total synthesis achieves calamistrin A and uvarigrin from tartaric acid, indicating potential in natural product chemistry.
The total synthesis of Annonaceae acetogenin natural products calamistrin A and uvarigrin was accomplished from the chiral pool of tartaric acid. Mukaiyama oxidative cyclization was used for the formation of trans-2,5-disubstituted tetrahydrofuran, while samarium-iodide-mediated deoxygenation of an α-hydroxy group in a masked α,β-dihydroxy ketone and Evans reduction were used for the installation of the trans-1,3-diol unit.
