The aim of this project is to improve (Refinement) animal models for virus-induced encephalitis and subsequent autoimmune responses. Enterovirus-induced encephalitis is used as an example, as it triggers a strong interferon response.Current models often rely on direct injection of the virus into the brain or brain ventricles of neonatal mice. While this approach takes advantage of the high susceptibility of young animals, it has disadvantages in terms of animal welfare. It does not reflect the natural route of infection and limits proper monitoring of animal burden as well as the use of analgesia.An alternative approach is therefore proposed: infection of adult mice via the intraperitoneal route. This leads to systemic viral spread followed by involvement of the central nervous system, which more closely reflects the natural course of infection in humans. This approach contributes to Refinement and may also support Reduction of animal use.
