As a core metabolic enzyme of the endocannabinoid system (ECS) and a key regulatory factor in lipid metabolic reprogramming, monoacylglycerol lipase (MAGL) exhibits a unique dual role in tumorigenesis and progression. This article systematically reviews the role of MAGL in tumors and provides a comprehensive summary of the development progress of MAGL inhibitors in tumors, including the structural characteristics, mechanisms of action, and pharmacological activities of irreversible and reversible inhibitors, with a focus on analyzing the advantages and limitations of various inhibitors. In addition, this paper also explores combination therapy based on MAGL, providing new ideas for tumor treatment. At the same time, the article prospects the application potential of MAGL as a clinical therapeutic target.
