Assesses FDEP-TP framework's procedural and translational capacities, indicating potential for improved metabolic interventions.
Submission package for Article A3 of the FDEP-TP trilogy (Lancet Diabetes & Endocrinology). Contains the Lancet-format manuscript (.docx and .pdf), Quarto source, cover letter, four display items (DI1-DI4) at 300 dpi, refs_A3.bib (26 cited references), the numerical ledger, the F1-F9 verification report, and the master ledger CSVs. Article A3 reports the procedural and translational layer of the FDEP-TP framework: the nine-label Periprandial Type Profile (PTP) per-analyte vocabulary, the eight-rule precedence hierarchy that aggregates per-analyte labels into Integrated Enteropancreatic Pattern (IEP) Types I-V with .i/.ii subtypes and a/b/c glycaemic-context suffixes, the sixteen-script reproducible analytic pipeline, and a hypothesis-generating clinical-decision algorithm for agonist selection (single GLP-1 RA, dual GIP/GLP-1, triple GLP-1/GIP/glucagon retatrutide as the Tipo VI extension) and metabolic-surgery indication. Pre-registered ecological proof-of-concept analysis (OSF DOI 10.17605/OSF.IO/3CZRE; v10.0 protocol frozen 2026-04-22): 23 source studies, 71 (Author x Cohort) tuples, 55 productive cohort-time-arms, 2,750 pseudo-subjects across 7 canonical cohorts and 11 analyte forms (master_table.csv SHA-256 2829cd78018e411783671ec00f849647858bda552cfa4ec23ad505ba9704a117). Three signatures: Type V bariatric-dominant (RYGBP 43.3%, SG 30.7%; Bonferroni q=0.035), Type III in obesity+T2DM (40.0%) and post-caloric-restriction (36.8%) (q=0.035), Type I.I in non-obese-without-T2D reference (32.2%). PTP per-analyte distribution across 8,050 classifications: 67.1% Preserved. Threshold sensitivity reassigns <=6.8%. The framework is hypothesis-generating; TRIPOD+AI declared pending prospective individual-participant validation. Companion paper A1 (conceptual-physiological layer) and A2 (mathematical-statistical layer; tag A2-submitted-2026-05-09) complete the trilogy. Supplementary S2 publishes the operational acquisition protocol that prospective validation studies should follow.
