ANP counteracts AngII-induced aldosterone secretion by activating PDE2A to degrade cAMP and prevent PKA activation.
Why the study?
It was not known whether cGMP-stimulated phosphodiesterase (PDE2A) participates in counteracting AngII-induced aldosterone secretion in adrenal zona glomerulosa cells.
Does Atrial natriuretic peptide (ANP) counteract acute Angiotensin II-induced aldosterone secretion via PDE2A in primary adrenal zona glomerulosa cells?

PDE2A knockdown in primary ZG cells may clarify ANP-AngII crosstalk; leaves open whether this informs aldosterone-targeted hypertension therapies.
BACKGROUND AND Atrial natriuretic peptide (ANP) prevents excessive aldosterone secretion from adrenal zona glomerulosa (ZG) cells. ANP reduces ACTH-stimulated aldosterone by enhancing cAMP degradation through cGMP-stimulated phosphodiesterase (PDE2A). It is not known whether PDE2A also participates in counteracting AngII-induced aldosterone secretion. EXPERIMENTAL APPROACH: In this study, we established CRISPR/Cas9 based knockdown of PDE2A in primary ZG cells and measured aldosterone release and cAMP/protein kinase A (PKA) activity using ELISA and Förster resonance energy transfer (FRET) biosensors, respectively. KEY CONCLUSION AND IMPLICATION: ANP/cGMP signalling effectively counteracts acute AngII-induced aldosterone secretion by activating PDE2A, which degrades AngII-stimulated cAMP and prevents PKA activation. This novel signalling pathway could be a target for new aldosterone-reducing drugs.

Mohagaonkar et al. (2026) studied Aldosterone secretion. Atrial natriuretic peptide (ANP) and PDE2A knockdown was evaluated on Aldosterone release and cAMP/protein kinase A (PKA) activity. Atrial natriuretic peptide effectively counteracts acute Angiotensin II-induced aldosterone secretion by activating PDE2A, which degrades cAMP and prevents PKA activation.
