In silico analysis predicts interaction of LINC01133 and miR-181A2HG affecting tumor pathways in TNBC.
Triple-negative breast cancer (TNBC) is the most aggressive molecular subtype of breast cancer, characterized by high migratory and invasive capacity, in which non-coding RNAs play a relevant regulatory role, although their mechanisms remain largely unknown. This study investigated, using in silico approaches, the possible interaction between LINC01133, an intergenic lncRNA with dual function and tumor context-dependent, and miR-181A2HG, a miRNA precursor of increasing oncological interest, in the human Hs578T TNBC cell line. RNA-Seq data comparing the parental cell line with the LINC01133 knockout cell line identified five differentially expressed DEGs related to miRNAs, including miR-181A2HG. These miRNAs/miRNAs DEGs were subjected to functional enrichment analysis using a miRNA background dataset derived from TCGA-BRCA biobank, showing a significant association with processes related to cell proliferation, apoptosis, inflammation, migration, and metastasis. In parallel, molecular docking was performed between predicted internal structural regions of LINC01133 and miR-181A2HG, demonstrating stable complexes formation involving loops 1 and complex 2/3 of LINC01133. The convergence among transcriptomic profiles, functional enrichment, and structural modeling supports the predicted hypothesis of a direct association between LINC01133 and miR-181A2HG, potentially implicated in modulating pathways associated with tumor progression in TNBC Hs578T cell line.
