Deep-UV resonance Raman spectroscopy (DUV-RRS) enables rapid, label-free quantification of mRNA directly in intact lipid nanoparticles. Without deformulation, seconds-scale acquisitions isolate adenine and guanine signatures with minimal lipid/water interference, yielding calibrated concentration estimates across buffers and formulations. A multipixel SNR strategy improves detection at low signal. This in situ assay offers a high-throughput readout for manufacturing and quality control of mRNA formulations.
