Summary: To study the role of androgens in the feedback regulation of gonadotropin secretion, we measured the effects of administration of dihydrotestosterone undecanoate (DHTU) and of spironolactone. Basal and LRH stimulated LH/FSH levels were determined in: A) Six eugonadal men, before and after six weeks' DHTU 120 mg/day. B) Six agonadal subjects after 12 weeks' DHTU 120 mg/day. The results of B were compared to those of C) Six agonadal subjects without sex steroid treatment. D) Six eugonadal subjects were studied before and after six weeks' administration of spironolactone. In the two groups of eugonadal subjects, administration of either dihydrotestosterone or spironolactone had no effect on basal and LRH-stimulated gonadotropin levels. A clear but modest suppression was observed in agonadal subjects. Possibly DHT exerts some suppressive effect on gonadotropin secretion in the absence of other testicular products (estradiol, testosterone, inhibin), known to play a role in the negative feedback regulation. From these data it seems unlikely that in the eugonadal male circulating DHT has an important role in the feedback regulation of gonadotropin secretion. Zusammenfassung: Androgene im Regelkreis der Gonadotropinausscheidung beim Mann: Wirkungen bei Anwendung von Dihydrotestosteron auf eugonadale und agonadale Personen sowie von Sprinolacton auf eugonadale Personen Um die Rolle der Androgene im Regelkreis der Gonadotropinsekretion zu studieren wurden die Wirkungen bei Anwendung von Dihydrotestosteronundecanat (DHTU) und Spironolacton gemessen. Bestimmt wurden die Basiswerte und die Werte nach Stimulation mit LRH von LH/FSH bei: A) Sechs eugonadalen Männern, vor und nach sechs Wochen DHTU 120 mg/Tag. B) Sechs agonadalen Männern nach 12 Wochen DHTU 120 mg/Tag. Die Ergebnisse von B wurden verglichen mit denen von C) Sechs agonadalen Personen ohne Sexualhormonbehandlung. D) Sechs eugonadale Personen wurden untersucht vor und nach sechswöchiger Behandlung mit Spironolacton. Bei den zwei Gruppen von eugonadalen Personen hatte die Behandlung jeweils mit Dihydrostestosteron oder mit Spironolacton keine Auswirkung auf den Gonadotropinspiegel basal und nach LRH-Stimulation. Eine deutliche aber bescheidene Hemmung wurde bei agonadalen Personen beobachtet. Möglicherweise zeigt DHT irgendeinen hemmenden Effekt auf die Gonadotropinausscheidung in Abwesenheit anderer testikulärer Produkte (Östradiol, Testosteron, Inhibin), die als Hemmfaktoren im Regelkreis gelten. Ausgehend von diesen Daten dürfte es unwahrscheinlich sein, daß beim eugonadalen Mann zirkulierendes DHT eine wichtige Rolle im Regelkreis der Gonadotropinsekretion hat.
