To determine whether sulfatides stimulate transmembrane signaling in human neutrophils through ligation of the cell-surface adhesion molecule L-selectin.
Treated human neutrophils with sulfatides or non-sulfated galactocerebrosides and measured cytosolic free calcium mobilization in the presence or absence of L-selectin shedding agents (chymotrypsin, PMA) and pertussis toxin.
Stimulated neutrophils with soluble or immobilized anti-L-selectin monoclonal antibodies (DREG-200, LAM1/3, LAM1/6, and LAM1/10) to verify receptor-specific calcium responses using fluorescence microscopy.
Measured tumor necrosis factor-alpha and interleukin-8 mRNA expression in response to sulfatide exposure before and after chymotrypsin-mediated L-selectin shedding.
Sulfatides induced an increase in cytosolic free calcium and elevated TNF-alpha and IL-8 mRNA levels in human neutrophils, whereas non-sulfated galactocerebrosides elicited no response.
Cleavage and shedding of L-selectin by chymotrypsin or PMA completely abolished neutrophil calcium mobilization and cytokine mRNA upregulation following sulfatide exposure.
Cross-linked or immobilized anti-L-selectin antibodies directly stimulated cytosolic free calcium influx, and neither antibody- nor sulfatide-induced signaling was blocked by pertussis toxin.
