To evaluate the effectiveness of single subcutaneous injections of specific pneumococcal capsular polysaccharides in preventing type-specific pneumococcal pneumonia and carriage.
Immunized alternate human subjects with a single subcutaneous injection containing 0.03 to 0.06 mg of capsular polysaccharides from pneumococcus types I, II, V, and VII.
Evaluated the onset and duration of immunity, incidence of pneumonia, and nasopharyngeal carrier rates in immunized versus non-immunized control individuals over at least 6 months.
Immunization conferred protection specifically against pneumococcus types I, II, V, and VII starting within 2 weeks and lasting at least 6 months, but did not protect against heterologous types.
Nasopharyngeal carrier rates for types I, II, V, and VII were significantly reduced in immunized subjects compared with controls.
Pneumonia incidence was substantially reduced among non-immunized subjects in the population, attributed to lower overall bacterial carrier rates.
