To evaluate whether osmotic-release methylphenidate results in a slower onset of presynaptic dopamine transporter blockade and decreased drug detection and likeability compared to immediate-release methylphenidate.
Randomized trial in 12 healthy adults administered single doses of immediate-release methylphenidate (40 mg) or osmotic-release methylphenidate (90 mg) on two separate occasions.
Plasma d-methylphenidate concentrations and subjective responses on a detection and likeability questionnaire were measured hourly for 10 hours post-dose.
Positron emission tomography with carbon-11-labeled Altropane quantified central dopamine transporter receptor occupancy at hours 1, 3, 5, and 7.
Osmotic-release methylphenidate produced a longer time to maximum plasma concentration and longer time to peak dopamine transporter occupancy than immediate-release methylphenidate, despite similar maximum concentration values.
Participants reported no drug detection or likeability with osmotic-release methylphenidate compared with immediate-release methylphenidate.
