Key result
Ataluren stabilizes muscle performance with no cardiac or respiratory decline in symptomatic Duchenne carriers.
Why the study?
Symptomatic DMD female carriers are not included in treatment indications such as ataluren, and little data documenting their management exists in the literature.
Does ataluren improve muscle strength, respiratory function, and cardiological function in symptomatic female carriers of Duchenne muscular dystrophy?
Case Report (n=4)
Yes
Does ataluren improve muscle strength, respiratory function, and cardiological function in symptomatic female carriers of Duchenne muscular dystrophy?
Ataluren therapy may provide temporary improvement or stabilization in muscle performance and maintain cardiac and respiratory parameters in symptomatic Duchenne muscular dystrophy female carriers.
Observed stabilization without cardiorespiratory decline in 4 carriers; hypothesis-generating, needs controlled trials before adoption.
carrier". Though there is no effective cure for DMD, therapies are available to slow the decline of muscle strength and delay the onset and progression of cardiac and respiratory impairment. These include ataluren for patients with nonsense mutations, and antisense oligonucleotides therapies, for patients with specific deletions. Symptomatic DMD female carriers are not included in these indications and little data documenting their management, often entrusted to the discretion of individual doctors, is present in the literature. In this article, we report the clinical and instrumental outcomes of four symptomatic DMD carriers, aged between 26 and 45 years, who were treated with ataluren for 21 to 73 months (average 47.3), and annually evaluated for muscle strength, respiratory and cardiological function. Two patients retain independent ambulation at ages 33 and 45, respectively. None of them developed respiratory involvement or cardiomyopathy. No clinical adverse effects or relevant abnormalities in routine laboratory values, were observed.

Dori et al. (2024) conducted a case report in Symptomatic Duchenne muscular dystrophy (DMD) carriers with nonsense mutations (n=4). Ataluren was evaluated on Change in 6-minute walk distance (6MWD). Ataluren treatment for up to 73 months in 4 symptomatic Duchenne carriers led to stabilization or temporary improvement in muscle performance, with no patients developing cardiac or respiratory decline.
