Deletion of type VIII collagen in mice significantly reduced systolic blood pressure to 87.0 mmHg compared to 103.0 mmHg in wildtype mice, and increased carotid artery functional distensibility.
Why the study?
Does the deletion of type VIII collagen affect elastin deposition and vascular smooth muscle cell phenotype?
Does the deletion of type VIII collagen affect elastin deposition and vascular smooth muscle cell phenotype?
Deletion of type VIII collagen promotes elastin deposition and a quiescent vascular smooth muscle cell phenotype, highlighting its role in vascular biology and potential impact on blood pressure and arterial distensibility.
studies revealed that the absence of Col-8 dramatically increased tropoelastin mRNA and elastic fiber deposition in the ECM, which was decreased with exogenous Col-8 treatment. These findings suggest a causative role for Col-8 in reducing mRNA levels of tropoelastin and the presence of elastic fibers in the matrix. Moreover, we also found that Col-8 and elastin have opposing effects on VSMC phenotype, the former promoting a synthetic phenotype, whereas the latter confers quiescence. These studies further our understanding of Col-8 function and open a promising new area of investigation related to elastin biology.

Mohabeer et al. (2021) studied Normal physiology / Col-8 deficiency. Deletion of type VIII collagen (Col8-/-) vs. Wildtype (Col8+/+) was evaluated on Systolic blood pressure (mmHg) (MD -16, p=0.00760). Deletion of type VIII collagen in mice significantly reduced systolic blood pressure to 87.0 mmHg compared to 103.0 mmHg in wildtype mice, and increased carotid artery functional distensibility.
