When sources of error of preparative and optical nature are avoided, a very high degree of accuracy can be attained by scanning photometry of Feulgen-stained and adequately flattened cell nuclei. In tetraploid rat liver nuclei, the spread observed in readings of different nuclei and of one nucleus in different positions to the optical axis is virtually the same, with a variation coefficient of below 1 %.
