Methods of breaking BP8 (C3H) ascites sarcoma cells have been compared with respect to the release from the cells of histocompatibility antigen as measured by haemagglutination inhibition. Hypotonic lysis of the cells liberates antigen, leaving the nuclei intact and easily sedimentable. A method is described for preparation in high yield of a highly active enriched fraction of the lysates. Adaptation of the method to ascites leukaemia cells is described. The products induce haemagglutinin formation when injected into allogeneic mice.
