Clofazimine plus moxifloxacin is linked to a ~13-ms increase in QTcF prolongation.
Why the study?
Does the combination of QT-prolonging drugs like clofazimine and moxifloxacin increase QT interval prolongation in children with rifampicin-resistant tuberculosis?

While overall risk of severe QT interval prolongation is low, combining clofazimine and moxifloxacin in children with RR-TB results in greater QT interval prolongation than using either drug alone.
May warrant closer QTc monitoring with clofazimine-moxifloxacin in children; leaves open regimen implications for prospective trials.
= 0.0166) compared to those when other MFX- or LFX-based regimens were used. In conclusion, we found a low risk of QTcF interval prolongation in children with RR-TB who received at least one QT-prolonging drug. Greater increases in maximum QTcF and ΔQTcF were observed when MFX and CFZ were used together. Future studies characterizing exposure-QTcF responses in children will be helpful to ensure safety with higher doses if required for effective treatment of RR-TB.

Ali et al. (2023) conducted a cohort in Rifampicin-resistant tuberculosis (n=88). Clofazimine and moxifloxacin combination vs. Single QT-prolonging agents or other combinations was evaluated on Change in QTcF (ΔQTcF) (13.0 ms increase, p=<0.001). The concomitant use of clofazimine and moxifloxacin was significantly associated with a 13.0-ms increase in the change in QTcF compared to baseline or other regimens (P<0.001).
