Randomized trial evaluates glycemic variability in type 2 diabetes, indicating significant benefits of iGlarLixi therapy.
Introduction and Objective: iGlarLixi is a once-daily, fixed-ratio combination of the basal insulin glargine 100 U/mL (Gla-100) and the glucagon-like peptide-1 receptor agonist (GLP-1 RA), lixisenatide (Lixi). The aim was to evaluate the impact of iGlarLixi on glycemic variability (GV) by assessing postprandial glucose (PPG) excursions and derived time in tight range (dTITR) in people with type 2 diabetes (PwT2D) from the global LixiLan clinical trial program. Methods: This participant-level exploratory analysis pooled data from the LixiLan-L, LixiLan-O and LixiLan-G trials; the time to end of treatment was 30, 30 and 26 weeks, respectively. Comparators for the studies were: Gla-100 (LixiLan-L), Gla-100 and Lixi (LixiLan-O) and daily- or weekly-GLP-1 RA therapy (LixiLan-G). Endpoints for this analysis were change in PPG excursions and dTITR. Results: In 2,403 participants, iGlarLixi significantly improved PPG excursions at 0.5 hours and 1 hour after a meal versus Gla-100 (p<0.0001 for both; Table). iGlarLixi also robustly reduced PPG excursions after 2 hours versus Gla-100 (p<0.0001) and GLP-1 RA (p=0.0004). Further, iGlarLixi significantly improved dTITR versus all comparators (p<0.0001 for all; Table). Conclusion: iGlarLixi demonstrated consistent improvements in PPG excursions and GV versus Gla-100 and GLP-1 RA therapies in PwT2D. Disclosure L. Czupryniak: Speaker's Bureau; Current; Abbott.
