Overview of pyrazolo[1,5-a]pyrimidines' synthesis, activities, and functional properties, indicating their potential use in drug development.
Pyrazolo[1,5‐ a ]pyrimidine derivatives are a substantial class of N ‐heterocyclic compounds that profoundly impact heterocyclic chemistry owing to their remarkable pharmacological and photophysical properties. Consequently, several researchers have developed unique synthetic methodologies for the creation and subsequent functionalization of this scaffold. These alterations enhance structural diversity and synergize innovative synthetic routes with the prospective applications of these compounds. This article offers an overview of current developments in the synthesis and reactions of various pyrazolo[1,5‐ a ]pyrimidines, as well as the application of synthetic methods to the FDA‐approved drugs, the ring's fluorogenic properties, and its structure‐activity relationships with its biological potential. Within this framework, chemists can select the approach that best suits their design. As a result, pyrazolo[1,5‐ a ]pyrimidines have become a desirable substitute due to their small size, which is based on [5,6]‐fused N‐heterocyclic systems, their effective synthetic approaches, their simple functionalization methods, and their fluorescence properties. It has been discovered that electron‐withdrawing groups at C‐3 enhance the scaffold's anticancer activity against both pim‐1 kinase and CDK2. Furthermore, cyclic aliphatic substituents bearing amino groups at C‐7 enhance their antiproliferative activity. Regarding antimicrobial activity, SAR investigations reveal that heterocyclic aromatic rings at C‐7 and halogenated phenyl substituents at C‐5 significantly enhance antibacterial activity.
