Establishes genetic transformation system in carrot, indicating potential for improved breeding techniques.
In order to establish an effective genetic transformation system of carrot, we firstly established an effective callus-inducible system of carrot by using hypocotyls and cotyledons as explants from 3 kinds of carrot (Daucus carota var. sativa) breeding lines. Hypocotyls and calli of carrot were infected with Agrobacterium tumefaciens LBA4404 harboring pBI121. By using X-Gluc staining, we confirmed the transient expression of the GUS gene. PCR analysis of regenerated plants confirmed that the GUS gene had been introduced into the plant genome. All of these results indicated that the effective genetic transformation system had been established.
