Evaluates miR-6809 and miR-4753 as diagnostic biomarkers in triple-negative breast cancer, highlighting their prognostic implications.
Background Triple-negative breast cancer (TNBC) remains lethal due to paucity of targetable markers. MicroRNAs are involved in TNBC pathogenesis; however, their clinical translation in TNBC remain limited. We evaluated miR-6809 and miR-4753 expression in TNBC tissues, their potential as diagnostic biomarkers, and their correlations with tumor-related data. Methods Thirty-seven TNBC and fifteen breast fibroadenoma (BFA) patients were recruited. Histological grading, immunohistochemistry, and RT-qPCR for miRNA profiling were conducted. Results miR-6809 and miR-4753 were downregulated in TNBC tissues compared to BFA. In TNBC, miR-6809 and miR-4753 were positively correlated. miR-6809 was negatively correlated with TNM staging and tumor grade. miR-4753 was inversely correlated with TNM staging and carcinoma in situ. Both miRNAs showed excellent diagnostic potential. miR-6809 was associated with TNBC risk in the multivariate analysis (OR = 0.0001, 95% CI = 0.00–0.232). Bioinformatics analysis revealed ADIPOR2, CD84, CD86, IL6R, PDGFA, and STAT5A, as integrated targets of both miRNAs, enriched in EGFR tyrosine kinase inhibitor resistance, Th17 cell differentiation, and JAK-STAT signaling. Conclusion This study highlights miR-6809 and miR-4753 as potential candidate biomarkers for TNBC. miR-6809 showed particular association with TNBC risk, while both miRNAs correlate with TNM staging, suggesting potential prognostic relevance. Further validation studies are required before clinical utility.
