Sotagliflozin mitigates doxorubicin-induced acute cardiotoxicity in mice by reducing apoptosis, inflammation, and mitochondrial dysfunction via the activation of the AMPK/mTOR pathway.
Sotagliflozin may attenuate doxorubicin cardiotoxicity in mice; hypothesis-generating for human translation and AMPK/mTOR mechanisms.
loading were reduced. Mitochondrial dysfunction was also improved, as indicated by increased JC-1 aggregates and Bcl-2/Bax ratios. Notably, SOTA activated 5' AMP-activated protein kinase (AMPK) and inhibited mTOR signaling while protecting from DOX-induced cardiomyopathy, suggesting SOTA as a promising therapeutic for heart failure treatment.

Liu et al. (2026) studied Doxorubicin-induced cardiomyopathy (n=18). Sotagliflozin vs. Doxorubicin alone and saline vehicle was evaluated on Left ventricular internal dimensions in diastole (LVIDd) and systole (LVIDs) (p=<0.001). Sotagliflozin significantly reduced doxorubicin-induced left ventricular enlargement (LVIDd, P<0.001) and mitigated apoptosis and mitochondrial dysfunction through the AMPK/mTOR pathway in mice.
