LncRNA Dancr overexpression delays myocardial fibrosis in AF models by sponging miR-146b-5p to promote Smad5.
Why the study?
Atrial fibrillation is closely associated with myocardial fibrosis, but the function of the lncRNA Dancr/miR-146b-5p/Smad5 axis in AF-related myocardial fibrosis was unknown.
Does lncRNA Dancr affect myocardial fibrosis in atrial fibrillation models via the miR-146b-5p/Smad5 axis?
Does lncRNA Dancr affect myocardial fibrosis in atrial fibrillation models via the miR-146b-5p/Smad5 axis?
LncRNA Dancr delays myocardial fibrosis in atrial fibrillation models by sponging miR-146b-5p to promote Smad5 expression, suggesting a potential therapeutic target for AF-related atrial remodeling.
May modulate MF in AF models via Dancr axis; extends mechanistic insights but leaves open in vivo validation.
Objectives: Atrial fibrillation (AF) is the most frequent arrhythmia, and myocardial fibrosis (MF) has a close association with atrial remodeling and leads to AF. This study aimed to explore the function of the long non-coding RNA (lncRNA) differentiation antagonizing non-protein coding RNA (Dancr)/microRNA (miR)-146b-5p/Smad5 axis on MF in AF mice. Methods: HL-1 cell rapid pacing models were constructed, and after lncRNA Dancr and miR-146b-5p-related construct transfection, cell viability and cell apoptosis were determined. Results: HL-1 cell rapid pacing models suggested that miR-146b-5p overexpression reversed the inhibitory effects of lncRNA Dancr overexpression on MF in HL-1 cells, and Smad5 interference reversed the ameliorative effects of miR-146b-5p interference on MF in HL-1 cells. Conclusions: LncRNA Dancr can sponge miR-146b-5p to promote Smad5 expression, thereby delaying MF in AF mice.
