We present a synergistic Jones-matrix mapping framework that integrates polarization interferometry and digital holography to uncover the hidden multifractal hierarchy of optical anisotropy in soft matter biological films. By leveraging original phase-scanning algorithms and digital holographic wave front reconstruction, this method enables the selective visualization of the real and imaginary components of Jones matrix operators, effectively decoupling latent birefringence and dichroism across varying phase depths. To quantify the complex spatial architectures of the specimens, a computational engine based on multifractal formalism and wavelet analysis was implemented. This analytical scheme allows for the characterization of self-similar supramolecular hierarchies that remain unresolved by conventional Stokes polarimetryю Experimental results demonstrate good preliminary discriminatory accuracy within the studied sample and confirm the diagnostic sensitivity of this method for identifying structural biomarkers of occult pathological changes. This is supported by analysis of various biological fluids, including blood samples from patients with pre-infarction and acute infarction, as well as from patients with other pathologies. The proposed 3D Jones-matrix imaging modality provides a robust technological platform for next-generation laser diagnostics, specifically aimed at the multiscale analysis of latent optical anisotropy in complex biopolymer systems.
